What It Is MOTS-C (Mitochondrial Open Reading Frame of the 12S rRNA-C) is a 16-amino acid mitochondria-derived peptide (MDP) encoded within the mitochondrial genome — specifically within the 12S ribosomal RNA gene. It is one of a growing class of mitochondria-derived peptides with broad systemic effects. MOTS-C is understood to translocate to the cell nucleus in response to metabolic stress, where it regulates nuclear gene expression related to metabolic homeostasis. Preclinical studies have demonstrated its role in enhancing insulin sensitivity, activating AMPK (AMP-activated protein kinase), promoting fatty acid oxidation, and improving physical endurance. Levels of MOTS-C are known to decline with age.
How It Is Used MOTS-C is reconstituted with bacteriostatic water and administered via subcutaneous injection in research protocols. It is used in metabolic disease modelling, mitochondrial biology research, insulin sensitisation studies, and ageing pathway investigations.
